Hammett lahir di San Francisco, California tanggal 18 November 1962, untuk Teofila (Oyao) dan Dennis L. Hammett, pelaut pedagang. Dia belajar gitar pada usia 15 dan bergabung Metallica untuk menggantikan Dave Mustaine pada tahun 1982. Dia telah memenangkan beberapa Grammy dengan Metallica dan berpartisipasi dalam class action law suit terhadap Napster. Ayahnya orang Irlandia, Inggris, Skotlandia, dan keturunan Jerman, dan keluarga ibunya adalah orang Filipina.
